Bisulfite modification of DNA is the most commonly used method for DNA methylation studies providing single nucleotide resolution. This technology is based on the chemical conversion of unmethylated cytosine to uracil. Methylated cytosines are protected from this conversion allowing to determine DNA methylation at the singe nucleotide level.

Due to the limited amount of DNA available, determination of methylation patterns in DNA from precious and limited sample materials e.g., formalin-fixed paraffin-embedded (FFPE) tissue or cfDNA is especially challenging. However, such sample types are of high interest for the successful identification of biomarkers.
